Summary:

This result consists on the first characterization of the influence of parameters like pH and ionic strengh from the environment on the structural stability of recombinant proteins such as:

  • The invertase expressed in Hansenula polymorpha and Pichia pastoris,
  • Aspartica protease and dextranase expressed in Pichia pastoris,
  • RHu-IFN-g (interferon g) and rSk (streptokinase) expressed in E. Coli.through the application of a combination of a group of physico-chemical methods (differential scanning calorimetry, circular dicroism, fluorescence and NMR) through which it is obtained the elucidation of its folding.

The combination of different methods allowed achieving novel information about:

  • a) the folding nature from some macromolecules and from the forces that influence the stability of these proteins on their native state,
  • b) the thermodynamic analysis of the interactions among macromolecules and specific ligands according to the temperature inside the study of the oligomerization process of proteins, or the evaluation of the influence of macromolecule stability inside a specific formulation.

The results obtained from this work have been published in 5 magazines of great impact.
