

The work deals with a new technology for the obtention of a Bromeline enzymatic preparation from pineapple sprouts. It was developed during 5 years by a group of researchers from different disciplines. The novelty is given by the design and optimization of an original technology which is efficient and has achieved good economic results, and where with the introduction of SH groups in the enzyme active center and the surveillance of other technological parameters it was possible to obtain better yields in terms of proteins and specific activities avoiding the proteolysis during the extraction.
This result has been acknoledged by the issuing of a patent by the OCPI. The work was completed with the purification of the enzyme by applying a novel combination of chromatographic means which allowed the molecular identification. The works were supported by essays and lab tests, as well as by a comprehensive bibliographic search, physical-chemical and lab characterization, toxicological analysis of protease, clinical studies to show its anti tumor activity, and stability, electrophoretic, scaling and application studies comprising therefore a wide range of the research, the innovation, the scaling and the application of the results.
Works presented have been part of a doctor and two master thesis and has given origin to the exhibition of the main results in 21 scientific events, three of them being very important internacional events on applied biotechnology, as well as 14 publications. They have received important awards: Mention at the 1997 National Science and Technology Forum , Outstanding Work at 12th National Forum for Students of Farming and Animal Husbandry Sciences, acknowledged by the Minister of MES on 1998, Health National Award on 2000.
The result was scaled and produced at the Biopreparations National Center (BIOCEN) at no extra cost but availing the installed industrial and technological capacities and 140 kg of the preparation have been produced, amount considered significant for being a product of high aggregate value and relatively limited application. The economic study carried out states that according to a comparison with the 2000 SIGMA catalogue and taking into account the yields obtained in the industrial process, the crude bromeline preparation allows to recover 17 164.55 USD a year, and the purified preparation allows to recover 587 153 USD a year, taking into consideration the import level of similar preparations.
The most important application has taking place in the biotechnology field where the crude preparation allowed the obtention of a Clorella Vulgaris hydrolyzed protein with a high nutritional value and multiple uses. The preparation had a confirmed anti-tumor activity in 5 of the 6 tumors chosen for the study and has a wide application in the treatment of leukemia P’338, Lewis lungs carcinoma, sarcoma S-37 and other tumors and so the result has a wide range of applications in medicine for the treatment of severe diseases. The work is guaranteed by the research, scaling and production centers engaged in its development.
